制备方法

1.以多孢木霉菌(Tolypocladium inflatum Cams)为生产菌株。75L罐内装50L种子培养基,接种5×109个孢子,Ph=5.4—4.3,培养72h,得到一级种子液。750L发酵罐内装500L发酵培养基,种入上述一级种子液,培养6天,得二级种子液。4500L发酵罐内装3000L发酵培养基,发酵12天,得到环孢素发酵毒。往发酵液中加人等体积的乙酸乙酯提取,分出有机层,减压蒸发,得粗品。融化处理,可得环孢素和环孢菌素C组分,其中环孢素的产量约为150~200mg/L环孢菌素C约为50~100mg/L。 2.以多孢木霉菌为生产菌株。先制备环孢素发酵液。向发酵液中加入乙酸乙酯提取,分出有机层,减压蒸发,得粗品。经纯化处理可得。

用途简介

优级纯。

用途

1.新型免疫抑制剂。为由十一个氨基酸组成的环状多肽物质。它抑制体液和细胞传递的免疫功能,而不抑制骨髓,不减少白细胞数。能阻止对移植组织或器官的排斥作用。临床用于肾移植,肝、肺移植及心脏移植,另外,对自身免疫疾病,以及血吸虫病、疟疾、糖尿病、艾滋病等有一定疗效,还有抗炎症性能。

2.治疗白血病、癌、肾移植、结核病等。

3.免疫抑制剂。由十一个氨基酸组成的环状多肽物质。可抑制淋巴细胞生成干扰素。它对网状内皮系统吞噬细胞无影响。环孢素不同于细胞毒类药物的作用,它仅抑制T细胞介导的细胞免疫而不致显著影响机体的一般防御能力。它抑制体液和细胞传递的免疫功能,而不抑制骨髓,不减少白细胞数。能阻止对移植组织或器官的排斥作用。另外,它对自身免疫疾病以及血吸虫病、疟疾、糖尿病、艾滋病等有一定
疗效,还有抗炎症性能。环孢素在临床上主要用于防止异体器官或骨髓移植时
排异等不利的免疫反应,常和糖皮质激素合用。
