2-氨基吡啶 制备方法及用途

制备方法

通常是在溶剂存在下,由氨基钠与吡啶反应。反应时逸出氢气,同时生成氨基吡啶的钠衍生物,后者经水解即生成游离的氨基吡啶。操作示例1将氨基钠加入干燥的甲苯中,加热回流,加吡啶同时通入氮气,继续回流6h。反应结束,冷却至20℃,通氮,缓缓加水,加毕,升温至60-70℃,静置分取甲苯层,水层保持60-70℃,用甲苯提取2次,减压回收甲苯,收集100-140℃(8kPa)馏出液,得2-氨基吡啶。收率为66-70%。操作示例2将甲苯加入干燥的不锈钢罐内,加热,蒸出甲苯直至馏出液为澄清。冷至35℃以下,加入氨基钠。升温至108℃,滴加吡啶。加毕,回流6h。冷至40℃加水,70℃搅拌1h。冷至30℃以下,分去水层(甲苯提取1次)。甲苯层用无水碳酸钠干燥。回收甲苯后,冷却固化得2-氨基吡啶。收率为65%。2-氨基吡啶的其他6-氨基烟酸在高于熔点的温度下熔融脱羧;由吡啶甲酰胺进行霍夫曼反应制取;通过2-溴吡啶氨解。工业生产中是向反应器中加入新制的氨基钠颗粒及甲苯,加热至110℃微有回流时,于搅拌下滴加吡啶,冷却控制反应速度,待反应缓慢后,继续加热搅拌回流3h;冷却至40℃加水分解,至反应物全部溶解时,趁热分去下层碱液,油层蒸去甲苯后,减压蒸馏,冷却后固化,得白色结晶,收率60%-63%。
