制备方法

方法1:1.42Gn-甲基-1-萘甲胺盐酸盐和2.89g碳酸钠溶于10ml二甲基甲酰胺中,在室温下滴加1.25g肉桂基氯。加毕,在室温下继续搅拌18h。过滤,滤液减压浓缩。剩余物溶于甲苯,无水硫酸钠干燥,过滤,浓缩,得萘替芬,沸点162~167℃/2.00Pa。萘替芬和含氯化氢的异丙醇溶液作用,可得盐酸萘替芬,熔点177℃(丙醇重结晶)。也可用异丙醇-乙醚重结晶。 方法2:以1-氯甲基萘为原料。在5~7℃下,往1-氯甲基萘的乙醇溶液中滴加30%甲胺乙醇溶液,加毕在室温搅拌过夜。蒸除乙醇,剩余物溶于氯仿,洗至中性,干燥,浓缩,减压蒸馏收集133~134℃/0.533kPa的馏分,即为N-甲基-1-萘甲胺,收率70%。在室温下将浓盐酸加到N-甲基-1-萘胺中,再加入碳酸钠和二甲基甲酰胺,滴加肉桂基氯,搅拌后过滤。滤液浓缩至干,加入甲苯,干燥,浓缩。再加入异丙醇,用浓盐酸调至Ph=3。加入乙醚,过滤,水洗,干燥,得白色固体的盐酸萘替芬,收率59.3%,熔点175~177℃。 方法3:在冰浴冷却下,向30%甲胺的乙醇溶液中,滴加1-氯甲基萘的无水乙醇溶液,然后在室温下放置过夜,可得N-甲基-1-萘甲胺。将其溶于乙醇,加入浓盐、35%甲醛水溶液和苯乙酮,加热回流,并补加聚甲醛细粉,进行Mannich反应得2-[N-甲基-N-(1-萘甲基)氨基]乙基苯基酮。在催化量相转移催化剂存在下,在乙酸乙酯中,用硼氢化钠把羰基还原为羟基。最后在盐酸中,回流脱水得萘替芬的盐酸盐。总收率30.1%。
