阿朴吗啡 制备方法及用途

制备方法

由吗啡经消除、重排、成盐而得。将吗啡加入磷酸中,水浴加热,内温不超过50℃。搅拌至全溶。冷却置于吸收罐内,通入氯化氢气体至反应液重达吗啡量的2.5倍。将其移入反应罐内,继续通入氯化氢气体,同时加热至124-126℃,保温反应3小时。将反应液放入冰水中,冷至20℃以下,搅拌下慢慢加入碳酸钠至胶状物析出完全为止。分取胶状物加入浓盐酸中,在沸水中溶解胶状物盐酸盐,再加入亚硫酸钠溶液中,用二氯乙烷-异丙醇混合溶媒提取2次。提取液于搅拌下加入浓盐酸,析出粗品。将所得粗品用浓盐酸浸泡、热蒸馏水溶解、加活性炭脱色、真空干燥等步骤便得到成品。
