杆菌肽锌 制备方法及用途

制备方法

生产菌是枯草杆菌。培养基是脱脂大豆粉(5%)、蔗糖(1.2%)、硫酸铵(0.2%)、碳酸钙(0.2%)等。菌的培养过程可分为菌的发育和抗菌素产生两个阶段。产生杆菌肽时的最适宜温度是37℃左右,而细菌发育期的温度则需略高1-2℃,每分钟的通气量为发酵液量的90%。产生的杆菌肽在pH值为4-5.7的条件下,吸附于羧酸型交换树脂上,然后用弱碱(氨)洗脱。由地衣形芽孢杆菌(Bacillus licheni formus)发酵而得。以黄豆粉,淀粉、玉米浸出物、糖蜜和无机盐为培养基,接种5%-10%,发酵30-40h。发酵7-8h后,PH从7.3-7.5降至6.06;发酵初期温度控制在30℃左右,经过2-3h温度提高到37℃。发酵结束后用浓盐酸调PH至5.5,然后在20M3发酵液中加入氧化锌50kg,混合30min。然后进行真空,蒸发浓缩,使干物质含量从4.5%-5%提高到17%-20%,再经喷雾干燥得产品。

合成制备方法
